Tissue clearing is one of the more efficient ways to facilitate 3D imaging of tissues, and hence generates massive volumes of complex data, which requires powerful computational hardware and software to store, process, analyze, and visualize.
3DISCO (stands for “3D imaging of solvent-cleared organs“) [1] is a histological method that make biological samples more transparent (so called “cleared”) by using series of organic solvents for matching the refractive index (RI) of the tissues with that of the surrounding medium.
Special tissue clearing techniques have been developed for this purpose, e.g. 3DISCO, CUBIC and CLARITY. Depending on the index of refraction of the cleared sample, matching immersion fluids and special long-distance objectives must be used during imaging.
