Construction on the expressway commenced in 1983, with the first two phases completed by 1988. This section involved the widening of several existing roads along the way, such as Ayer Rajah Road and Upper Ayer Rajah Road, as well as the construction of what was then the longest road viaduct, the Keppel Viaduct, from where the eastern end of the expressway commences.
The latest expressway completed is the 5 km Marina Coastal Expressway (MCE), which links the East Coast Parkway and Kallang-Paya Lebar Expressway to Marina South and Ayer Rajah Expressway and includes Singapore's first undersea tunnel. Construction started in 2008 and the expressway opened to traffic 29 December 2013.

Travelling from west to east in a clockwise direction, the route begins at Queenstown with a connection to the Ayer Rajah Expressway. The route continues northward through the residential areas of Bukit Timah via Farrer Road and Adam Road, with the first connection to the Pan Island Expressway .

The 5-kilometre (3.1-mile) MCE connects with the southern end of the Kallang–Paya Lebar Expressway (KPE) and its junction with the East Coast Parkway (ECP) to the eastern end of the Ayer Rajah Expressway (AYE). This links the eastern and western parts of Singapore to the New Downtown, currently being developed in the Marina Bay area.
It will be rolled out to Central Expressway (both blue screens and tunnels), East Coast Parkway, and Bukit Timah Expressway/Seletar Expressway that is near to the Woodlands Checkpoint, which was completed in October 2014. The second phase will be rolled out to Ayer Rajah Expressway and Kranji Expressway, which was completed in August 2015.
In 1998, Ayer Rajah Expressway was further expanded into Tuas. The extension followed the alignment of Jalan Ahmad Ibrahim. The road was widened, with all roundabouts replaced with flyovers, and interchanges and slip roads built along some parts of the expressway to serve the existing establishments along the road.
