Founded in 1956, KSMC is the oldest medical city in the Kingdom. It was inaugurated by King Saud bin Abdulaziz as the King Saud Medical Complex, and was at the time a 500-bed facility housing an emergency department and divisions in orthopedics, general surgery, obstetrics and gynecology, pediatrics, and a radiology department, along with a laboratory, pharmacy, and immunization department.
The King Saud Bin Abdulaziz University for Health Sciences was formally established in 2005 as a result of the positive feedback from the postgraduate programs in various medical fields being offered by the National Guard Health Affairs since the mid-1980s. It is now the best university in Saudi Arabia to provide health staff.

The advanced medical facilities at the King Abdulaziz Medical Cities in Riyadh, Jeddah and Al-Ahsa proved to be critical foundations for the successful inception of the university. KSAU-HS's main campus is in Riyadh with two additional campuses in Jeddah and Al-Ahsa.

The College of Medicine in King Saud University was established as the first medical college in the Kingdom in 1967. One year later, King Abdul-Aziz University Hospital, became affiliated with the college and subsequently in 1981, King Khalid University Hospital and the new college building were established to become the main teaching campus and patient service facility.
