A diffusive barrier is between the chloroplasts (which contain RuBisCO) and the cytosol. This enables a bundle-sheath-type area and a mesophyll-type area to be established within a single cell. Although this does allow a limited C 4 cycle to operate, it is relatively inefficient.
Palisade mesophyll cells can contain 30–70 chloroplasts per cell, while stomatal guard cells contain only around 8–15 per cell, as well as much less chlorophyll. Chloroplasts can also be found in the bundle sheath cells of a leaf, especially in C 4 plants, which carry out the Calvin cycle in their bundle sheath cells.
The pores or stomata of the epidermis open into substomatal chambers, which are connected to the intercellular air spaces between the spongy and palisade mesophyll cell, so that oxygen, carbon dioxide and water vapor can diffuse into and out of the leaf and access the mesophyll cells during respiration, photosynthesis and transpiration.

[2] These layers are called the palisade parenchyma and spongy mesophyll. Palisade parenchyma cells can be either cuboidal or elongated.
The surface of the leaf is coated with a water-resistant waxy cuticle that protects the leaf from excessive evaporation of water and decreases the absorption of ultraviolet or blue light to minimize heating. The transparent epidermis layer allows light to pass through to the palisade mesophyll cells where most of the photosynthesis takes place.

This means water moves up the vessels. The last stage in the transpiration stream is the water moving into the leaves, and then the actual transpiration. First, the water moves into the mesophyll cells from the top of the xylem vessels. Then the water evaporates out of the cells into the spaces between the cells in the leaf.
