The COSMIC (Catalogue of Somatic Mutations in Cancer) database was designed to collect and display information on somatic mutations in cancer. It was launched in 2004, with data from just four genes, HRAS, KRAS2, NRAS and BRAF. [6] These four genes are known to be somatically mutated in cancer. Since its creation, the database has expanded rapidly.
The Cancer Genome Atlas (TCGA) is a project to catalogue the genomic alterations responsible for cancer using genome sequencing and bioinformatics. [1] [2] The overarching goal was to apply high-throughput genome analysis techniques to improve the ability to diagnose, treat, and prevent cancer through a better understanding of the genetic basis of the disease.
MMHCdb allows researchers to search for PDX models by such parameters as cancer site, diagnosis, genomic properties of the engrafted human tumor, and model treatment response. Fig. 3 shows the output of a search for PDX models whose engrafted tumors have amplified KRAS, the TP53 A159V mutation, a deletion of the ALB gene, and high KIT expression.
The mutation catalog of the tumor is compared to a reference mutation catalogue, or mutational signatures reference dataset, such as the 21 Signatures of Mutational Processes in Human Cancer [4] from the Catalogue of Somatic Mutation In Cancer database. [1]
The project operates within the scope of the International Cancer Genome Consortium, working with the other participating organizations and countries to build a database of genomic changes present in different types of cancer.
