Its southern terminus is in Gulfport, Mississippi, at an intersection with US 90. US 49 is approximately 516 miles (830 km) in length.

U.S. Route 49, a four-lane divided highway, follows the western border of the town and leads northwest 52 miles (84 km) to Jackson, the state capital, and 38 miles (61 km) southeast to Hattiesburg. Mississippi Highway 35 passes through the southern end of town and leads northeast 10 miles (16 km) to Mize and southwest 39 miles (63 km) to Columbia.
Hattiesburg gained its nickname, the Hub City, in 1912 as a result of a contest in a local newspaper. It was named because it was at the intersection of a number of important rail lines. Later U.S. Highway 49, U.S. Highway 98 and U.S. Highway 11, and later, Interstate 59 also intersected in and near Hattiesburg.
