The St Luke's Vestry was incorporated as local government body and appointed one member to the Metropolitan Board of Works. In 1867 a new vestry hall was completed at a cost of £6,400 (equivalent to £715,801 in 2023) on the St Luke's Workhouse site in the parish of St Leonard Shoreditch, fronting City Road.

James Brooks FRIBA (1825–1901) was an influential English Gothic Revival architect and designer. Brooks established his reputation through a series of landmark churches built in the East End of London in the 1860 and 1870s, and was awarded the Royal Institute of British Architects' Royal Gold Medal in 1895.
St Mark's also founded mission churches at St Matthew's, Custom House in 1860, which was later transferred to St Luke's Church, Canning Town before becoming a parish of its own in 1920. St Luke's itself was originally a mission church of St Mark's before being made a parish of its own on its permanent church's completion in 1875.

In 1862 St Leonard's parish was again divided to create the parish of "Shoreditch, St Michael". In 1866 a new parish of "Holy Trinity, Shoreditch" was formed from part of Saint Leonard's and part of neighbouring parish of Bethnal Green. In 1872 the parish of "Shoreditch, St Agatha" was created from part of Saint James, Curtain Road.
