IL-1ra functions as a competitive inhibitor of IL-1 receptor in vivo and in vitro. It counteracts the effects of both IL-1α and IL-1β. Upon binding of IL-1ra, the IL-1 receptor does not transmit a signal to the cell. IL-1ra inhibits the release of both IL-1α and IL-1β, IL-2 secretion, cell surface IL-2 receptor expression.
An inflammatory cytokine is a type of cytokine (a signaling molecule) that is secreted from immune cells and certain other cell types that promotes inflammation. Inflammatory cytokines are predominantly produced by T helper cells ( T h ) and macrophages and involved in the upregulation of inflammatory reactions. [ 1 ]
Interleukin 10 (IL-10) is a protein that inhibits the synthesis of a number of cytokines, including IFN-gamma, IL-2, IL-3, TNF, and GM-CSF produced by activated macrophages and by helper T cells. In structure, IL-10 is a protein of about 160 amino acids that contains four conserved cysteines involved in disulphide bonds. [33]
The protein encoded by this gene is a decoy receptor for certain cytokines that belongs to the interleukin-1 receptor family. This protein binds interleukin-1α (IL1A), interleukin-1β (IL1B), and interleukin 1 receptor antagonist (IL1Ra), preventing them from binding to their regular receptors and thereby inhibiting the transduction of their signaling.
IL-1R signalization activates immune response by activation of transcription of IL-1 target genes such as IL-6, IL-8, MCP-1, COX-2, IκBα, IL-1α, IL-1β, MKP-1. Components of signalization pathway of IL-1R which are involved in cellular response to IL-1 also mediate responses to other cytokines ( IL-18 and IL-33 ), Toll-like receptors (TLRs ...
IL-1R Signaling pathway. Interleukin-1 receptors (IL-1Rs) are cytokine receptors that transduce an intracellular signaling cascade in response to the binding of the inflammatory cytokine interleukin-1 (IL-1). This signaling cascade results in the initiation of transcription of certain genes involved in inflammation.
Caspase-1 Zymogen. Caspase-1/Interleukin-1 converting enzyme (ICE) is an evolutionarily conserved enzyme that proteolytically cleaves other proteins, such as the precursors of the inflammatory cytokines interleukin 1β and interleukin 18 as well as the pyroptosis inducer Gasdermin D, into active mature peptides.
