U.S. Route 27 (US 27) in Kentucky runs 201.120 miles (323.671 km) from the Tennessee border to the Ohio border at Cincinnati.It crosses into the state in the Lake Cumberland area, passing near or through many small towns, including Somerset, Stanford, and Nicholasville.
The highway crosses Mill Creek and intersects US 27, which bypasses Cynthiana to the west, shortly before entering the city. KY 356 enters the city along Pleasant Street and reaches its eastern terminus as the west leg of a five-legged intersection on the west side of the South Fork of the Licking River.

Cynthiana is located in southern Harrison County. U.S. Routes 27 and 62 pass through the city, passing west of the downtown area. US 27 leads north 56 miles (90 km) to Cincinnati and south 14 miles (23 km) to Paris, while US 62 leads northeast 46 miles (74 km) to Maysville and southwest 21 miles (34 km) to Georgetown.
In Harrison County, US 62 previously followed a route east of Cynthiana to start its concurrency with US 68 just south of Blue Lick State Park until it was rerouted to its current course to Maysville prior to 1940.
