State Route 3 (SR 3) is a 120.67-mile-long (194.20 km) state highway located in southern Maine. It is a major interregional highway, connecting the Interstate 95 corridor to the Atlantic coast . The western terminus is at SR 8 , SR 11 and SR 27 in Augusta and the eastern terminus is at SR 102 and SR 198 in Mount Desert .

Twitchell Brook enters the lake near a point where the Appalachian Trail provides a view of the lake, but there is no inlet. Road access to the lake is provided by Maine State Route 11 on the east side. This runs past Partridge Cove at the southeast corner of South Twin Lake and then serves the area up to Norcross.

State Route 203 (SR 203) was a part of the Maine system of numbered state highways, located in Waldo County. The route was 4.62 miles (7.44 km) long and ran from SR 131 in Waldo, to SR 139 in Brooks. It was the only state-numbered highway in Maine that was entirely locally maintained. As of 2023, State Route 203 no longer exists.
