St. Mary Church was built of limestone in a Romanesque Revival style. The façade is framed by two hexagonal towers; the spire of the north tower houses the bell, and is raised six feet (1.8 meters) above the south tower. The large central double entrance doors and smaller flanking doors are topped by an arched wooden infill panel.

Located on South Claiborne Street in downtown, it is bounded by Dauphin Street on the north, Franklin Street on the west, and Conti Street on the south. The front of the church faces east, toward the Mobile River, and overlooks Cathedral Square. The building, laid out in an east–west axis, is 164 ft (50 m) long and 90 ft (27 m) wide.
