Tegafur/uracil (abbreviation: UFT [1]) is a chemotherapy drug combination used in the treatment of cancer, primarily bowel cancer. UFT is an oral formulation combining uracil (a competitive inhibitor of dihydropyrimidine dehydrogenase ), and tegafur (a bioavailable 5-fluorouracil (5-FU) prodrug ) in a 4:1 molar ratio.
Tegafur is a chemotherapeutic prodrug of 5-fluorouracil (5-FU) used in the treatment of cancers. It is a component of the combination drug tegafur/uracil . When metabolised, it becomes 5-FU.
Tegafur/gimeracil/oteracil, sold under the brand name Teysuno among others is a fixed-dose combination medication used for the treatment of advanced gastric cancer when used in combination with cisplatin, [3] and also for the treatment of head and neck cancer, colorectal cancer, non–small-cell lung, breast, pancreatic, and biliary tract cancers.
Uracil is also involved in the biosynthesis of polysaccharides and the transportation of sugars containing aldehydes. [22] Uracil is important for the detoxification of many carcinogens, for instance those found in tobacco smoke. [23] Uracil is also required to detoxify many drugs such as cannabinoids (THC) [24] and morphine (opioids). [25]
This compound exists in all living organisms and can become part of DNA in both prokaryotic and eukaryotic cells through two mechanisms. The first is the removal of an amino group from cytosine to result in uracil and the second is the non-intentional incorporation of pyrimidine where thymine belongs in the DNA, resulting in dUMP. [3]
The viral polymerase incorporates these compounds with non-canonical bases. These compounds are activated in the cells by being converted into nucleotides; they are administered as nucleosides as charged nucleotides cannot easily cross cell membranes. [citation needed] At least one set of new base pairs has been announced as of May 2014. [15]
The ammonium can then enter the urea cycle which occurs in the cytosol and the mitochondria of cells. [5] Pyrimidine bases can also be salvaged. For example, the uracil base can be combined with ribose-1-phosphate to create uridine monophosphate or UMP. A similar reaction can also be done with thymine and deoxyribose-1-phosphate. [8]
Uramustine or uracil mustard is a chemotherapy drug which belongs to the class of alkylating agents. [1] It is used in lymphatic malignancies such as non-Hodgkin's lymphoma . It works by damaging DNA , primarily in cancer cells that preferentially take up the uracil due to their need to make nucleic acids during their rapid cycles of cell ...
