After refining their understanding of PPARα’s effects on NRAS MUT melanomas, the researchers capitalized on this finding by using a combination treatment of two compounds, fenofibrate (which activates PPARα) and DHA (also known as Omega-3), to successfully induced cell death in melanomas that were known to resist treatment with MAPK inhibitors.

Among atypical adverse effects, myositis can occur in patients under gemfibrozil therapy, especially in those with renal insufficiency or under co-treatment with statins. In contrast, the risk of myopathy is much lower if fenofibrate is used in replacement of gemfibrozil owing to its different pharmacokinetic pathway.
Consequently, these drugs have been used for the treatment of hypercholesterolemia and dyslipidemia. [ 4 ] [ 5 ] [ 6 ] Use of these agents as hypolipidemic agents has decreased markedly since the introduction of the statins , which are more efficacious than bile acid sequestrants at lowering LDL .
Most fibrates can cause mild stomach upset and myopathy (muscle pain with CPK elevations). Fibrates decrease the synthesis of bile acid by down-regulation of cholesterol 7 alpha-hydroxylase and sterol 27-hydroxylase expression, therefore making it easier for cholesterol to precipitate and increasing the risk for gallstones.
