Its western terminus is at ND 14 about 3.3 miles (5.3 km) south of the Canadian border and about 3.7 miles (6.0 km) north of Carbury. Shortly after the route begins, ND 43 enters Turtle Mountain State Forest , where the highway curves slightly but eventually straightens out again.
The first city on its route is Bowman at the junction of US 12. Continuing north, it passes between North Dakota's two highest points, White Butte and Black Butte. Near Amidon US 85 heads east for nine miles (14 km) before going back north along the Little Missouri National Grassland for about 125 miles (201 km).

ND 24 begins at an intersection with ND 6 east of Selfridge, just north of the South Dakota–North Dakota state line. From there, it travels east until it meets ND 1806 south of Fort Yates. From there, the two routes run north concurrently, through Fort Yates and alongside the west bank of the Missouri River as part of the Lewis and Clark Trail.
