[2] U.S. Route 9W passes through the eastern part of the town. US Route 44 and NY 55 cross the northwestern corner of the town. The town was named after the Duke of Marlborough.
Marlborough is drained by Minnewawa Brook, a westward-flowing tributary of the Ashuelot River and part of the Connecticut River watershed. The town's highest point is near its southeastern corner, on the side of Bigelow Hill, where the elevation reaches 1,516 feet (462 m) above sea level. Marlborough is served by state routes 101 and 124.
